【Animal Modeling】-Effect of Guiji Cream on fibronectin and basic fibroblast growth factor in a rat model of stage III pressure ulcer

  OBJECTIVE: The purpose of this study was to investigate the effect of Guiji cream on the levels of fibronectin and basic fibroblast growth factor in pressure ulcer granulation tissue in pressure ulcer model rats.

  METHODS: Among 128 SD rats, 32 were randomly selected as the sham-operated group, and the other 96 rats were implanted under the bilateral gluteal muscles of the rats, and the rats were established by the method of circulatory compression of ischemia-reperfusion. The stage III pressure ulcer model was randomly divided into model group, positive control group, and rose and cream group after 4 days. The sham operation group and model group were externally applied with normal saline gauze, and the positive control group was externally covered with hydrocolloid dressing. Topical application of rose and cream gauze to the sore site. At the end of the treatment, the effective time of the pressure ulcer wound, the healing speed and the healing rate of the pressure ulcer were counted, and the serum levels of the tail vein blood of the rats were detected on the 3rd, 7th, 14th, and 21st days of treatment respectively. The levels of inflammatory mediators interleukin-1β (interleukin-1β, IL-1β), interleukin-6 (IL-6) and C-reactive protein (CRP) were measured by solid-phase sandwich enzyme-linked immunosorbent assay. The levels of FN and BFGF in pressure ulcer granulation tissue were detected by adsorption experiment.

  Results: Compared with the positive control group, the marked effect time and healing time of the Guiji cream group were shorter than those of the positive control group, and the wound healing rate and healing speed were higher than those of the positive control group, and the difference between the two groups was significant (P<0.05). At 28 days, venous blood CRP and IL-1β, IL-6 in the Guiji cream group were significantly lower than those in the model group and the positive control group (P<0.05), while BFGF and FN were significantly higher than those in the model group and the positive control group Concurrent level (P<0.05).

  CONCLUSION: Guiji cream may reduce wound injury by inhibiting the inflammatory response caused by inflammatory mediators IL-6 and IL-1β in pressure ulcer wounds, and increase the synthesis of BFGF and FN to promote the proliferation of interstitial cells and the synthesis of extracellular matrix. , thereby promoting the regeneration of wound granulation tissue and accelerating wound tissue repair.
