【Animal modeling】-Evaluation of the curative effect of Fenbid on the model of lumbar disc herniation in rats

  Objective To study the effect of cyclooxygenase inhibitor Fenbid on lumbar disc herniation.

  Methods Rats were modeled by compressing the dorsal root ganglion and transplanting autologous caudal nucleus pulposus to the lumbar spine. The COX (cyclooxygenase) inhibitor Fenbid was administered at a dose of 0.051 g/kg for 28 consecutive days. The motor function score, mechanical stimulation retraction response measurement, animal thigh circumference measurement and gait measurement were performed on the animals. After 28 days of administration, the lumbar vertebrae (L4-L6) and their nerves were fixed with 4% formaldehyde solution for histopathological examination, and serum and nerve root lesions were collected to determine the concentration of inflammatory factors.

  Results The COX inhibitor Fenbid could improve the animal motor function score, 50% mechanical foot withdrawal reflex threshold, thigh circumference ratio (modeling side/contralateral), sciatic nerve function index (SFI), dorsal root ganglion And the pathological changes of intervertebral disc, reduce the levels of inflammatory factors (PGE2, COX-2, NO, IL-1β, IL-6) in serum and lesions.

  Conclusion COX inhibitors can improve the symptoms of lumbar disc herniation model in rats.
