Objective To establish a stable and efficient rat model of hyperuricemia complicated with liver and kidney injury and gouty arthritis.
Methods Forty-eight male Sprague-Dawley rats (hereinafter referred to as SD rats) were randomly divided into 6 groups (8 rats in each group), which were blank control group (Control group), potassium oxonate group (OA group), oxygen Potassium oxonate + hypoxanthine group (OA+H group), potassium oxonate + yeast paste group (OA+YE group), 10% fructose + 0.2% ethambutol group (10% Fru + 0.2% EMB group) ), 2% potassium oxonate combined with 12% yeast special diet group (OA PO group) five different drugs and different concentrations of experiments, after 5 weeks, the serum uric acid, creatinine, urea and other biochemical indicators were measured and renal and Joint synovial muscle pathological section observation.
Results Compared with the blank control group, the serum uric acid level in the OA PO group was increased, accompanied by severe liver and kidney damage and joint inflammation. The OA, OA+H, OA+YE, 10%Fru+0.2% EMB groups did not reach high uric acid levels, but had liver and kidney damage and joint inflammation to a certain extent.
Conclusion The mixed diet of 2% potassium oxonate and 12% yeast extract has the best effect in establishing hyperuricemia and complications model, and this model is also suitable for the related research on renal damage and joint inflammation caused by hyperuricemia.
