Objective To dynamically observe the changes of pain emotion in chronic inflammatory pain model rats induced by complete Freund's adjuvant (CFA), and to explore the time differences of anxiety and depression in chronic inflammatory pain model rats.
Methods All experimental rats were randomly divided into blank control group and model control group. The model control group was injected with CFA into the foot to establish a chronic inflammatory pain model. Dynamically observe the changes of mechanical foot withdrawal thresholds (PWTs) in each group before modeling and 1, 3, 7, 18 and 26 days after modeling; observe the open field test (OF) at 28 days after modeling, and the elevated O maze at 29 days after modeling. Experiment (EZM), 30 d novelty food inhibition experiment (NSF), 12, 20 and 31 d after modeling marble burial experiment (MBT), 17, 25 and 35 d after modeling, sugar water preference experiment (SPT) in rats Behavior change situations.
Results One day after modeling, the PWTs of the model control group were significantly decreased (P < 0. 01), and were significantly lower than those of the blank control group during the whole experimental process (P < 0. 01). Compared with the blank control group, the central area movement distance, central area stay time and central area entry times of the model control group were significantly reduced in OF (P < 0.01); in EZM, the open arm movement of the rats in the blank control group was significantly reduced (P < 0.01). The distance, open arm dwell time, and open arm entry times were significantly decreased (P < 0. 05); in NSF compared with the blank control group, the feeding latency was significantly increased (P < 0. 05), and there was no significant difference in food consumption (P < 0. 05). P > 0. 05); and 31 d after MBT only, the number of buried marbles in the model control group was significantly increased compared with that in the blank control group (P < 0. 05); the model control group and the blank control group at each time point after SPT were modeled. There was no significant difference between the groups of rats (P > 0.05).
Conclusion CFA-induced chronic inflammatory pain model rats can show a variety of animal behavior changes related to anxiety-like emotions after 4 weeks, but no behavioral changes related to depression-like emotions were observed within 5 weeks.
