Objective To create a self-made restraint device for mice, and to establish a postnatal mouse restraint stress model for related stress experiments.
Methods Select a centrifuge tube with a size suitable for the size of the mouse, and prepare a simple mouse restraint device. On the second day after the mouse is born, the device is used to fix the postpartum female mouse in the device to restrict its movement. On the basis of separation of mother and child, daily The postpartum mouse restraint stress model was prepared by giving the female mice 3 hours of restraint stress and continuous modeling for 21 days.
Results In this postnatal mouse restraint stress model, the expression of ghrelin in the stomach, hypothalamus and serum was increased, the expression of 5-HT2 receptor and other genes was decreased, and there were depression-like symptoms such as decreased exercise and growth retardation of offspring.
Conclusion This article introduces the postnatal mouse restraint stress model. After many experiments, it has been proved that the modeling results have good stability and repeatability, which can be used for experimental research on restraint stress in mice.
