OBJECTIVE: To construct a rat model of atherosclerosis combined with disease and syndrome, and to provide an evaluation tool for the pharmacodynamics study of the corresponding traditional Chinese medicine.
Methods: Referring to relevant data and literature research, the diagnosis and treatment indicators for evaluating the rat model of atherosclerosis (Qi stagnation and blood stasis syndrome) were extracted and proposed, and Simvastatin Tablets and Tongmai Granules were used as counter-evidence drugs. Combined with the etiology of atherosclerosis For the modern research on the evolution law of pathogenesis and pathogenesis, the combined induction method of ice-water bath, gavage of fat emulsion and tail vein injection of bovine serum albumin is proposed to prepare atherosclerosis (qi stagnation and blood stasis syndrome) rat model of combination of disease and syndrome. During the experiment, the symptoms and signs of the rats in each group were investigated and scored. After 8 weeks of modeling, the four levels of blood lipids and blood coagulation, and the pathological changes of the ascending aorta were detected in the rats of each group for pre-testing of the modeling. After the model was successfully established, the rats were intervened with anti-evidence drugs, and the four levels of blood lipids and coagulation in the rats were measured, and the pathological changes of the ascending aorta and liver tissue were detected.
Results: After 21 weeks of modeling, compared with the normal group, the symptoms and signs of the rats in the model group changed significantly, the blood lipid level and fibrinogen content were significantly increased, and the activated partial prothrombin time was significantly decreased. In the ascending aorta Lipid deposition appeared under the membrane, and vacuoles and degeneration of hepatocytes appeared, basically in line with the clinical manifestations of atherosclerosis (Qi stagnation and blood stasis syndrome). The fibrinogen content was significantly decreased, and the intima of the ascending aorta and liver tissue were not abnormal.
CONCLUSION: The combined induction method of ice-water bath, gavage of fat emulsion and tail vein injection of bovine serum albumin for continuous 21 weeks can establish a rat model that simulates clinical atherosclerosis (Qi stagnation and blood stasis syndrome), and has a good success rate. high, similar to clinical symptoms and signs, which will provide an evaluation for the pharmacodynamics study of traditional Chinese medicine compounds against atherosclerosis (qi stagnation and blood stasis syndrome).
