Objective: To study the expression of serum cytokines in collagen-induced rheumatoid arthritis, cold-damp bi and damp-heat bi disease/syndrome model rats.
Methods: The rheumatoid arthritis model was established by using bovine type Ⅱ collagen and Freund's complete adjuvant. On this basis, cold-dampness and damp-heat stimulation were added to establish the combination model of cold-dampness arthralgia and damp-heat arthralgia. The serum was detected by microarray antibody chip. Cytokine expression.
Results: Compared with the normal group, activinA, CNTF, IFN-γ, TNF-α were up-regulated, MIP-3α, MMP-8 were down-regulated in RA group; CINC-2α, GM-CSF, IFN-γ were up-regulated, MIP- 3α was down-regulated; ICAM-1 was up-regulated, IFN-γ, MIP-3α, and RAGE were down-regulated in damp-heat bi group. Compared with RA group, β-NGF, CINC-2α, MMP-8, PDGF-AA were up-regulated, activin, activin and TNF-α was down-regulated; MMP-8 was up-regulated in damp-heat bi group, activin, CNTF, IFN-γ, IL-1α, IL-6, MIP-3α, RAGE, TNF-α were down-regulated. Compared with the paralysis group, CINC-2α, IFN-γ, IL-1α, MIP-3α, PDGF-AA, and RAGE were all up-regulated. Compared with the normal group, the ratio of IFN-γ/IL-4 in the RA group and the cold-damp paralysis group was increased. Compared with the RA group, the ratio of the cold-damp bi group was increased, and the ratio of the damp-heat bi group was significantly decreased.
Conclusion: Disease/syndrome factors have obvious effects on the expression profile of serum cytokines in rats, and there are significant differences among the models.
