【Animal Modeling】-Establishment and evaluation of a rat model of chronic salpingitis due to qi stagnation and blood stasis

Objective: To explore the construction method of chronic salpingitis model of qi stagnation and blood stasis type, investigate its feasibility and make a preliminary evaluation of the model, so as to lay a foundation for further experimental research.

Methods: SD rats were selected, and unpredictable repeated chronic stress stimulation (acoustic, light, electric, tail-clamping stimulation) and tubal surgery were used to construct a chronic tubal inflammation model of qi stagnation and blood stasis, and the symptoms were evaluated. The establishment of an animal model of chronic fallopian tube inflammation of Qi stagnation and blood stasis type was preliminarily evaluated.

Results: Compared with the model group, the total symptom score of the composite model group increased significantly (P<0.01). In the histomorphological observation, 80% of the fallopian tubes had severe vascular swelling, and 60% had severe tube wall thickening; In the model group, more inflammatory cells infiltrated in the fallopian tube lumen, and the mucosal epithelial cells fell off, accompanied by a large number of inflammatory cells around the fallopian tube. The index and aggregation index increased significantly (P < 0.01).

Conclusion: Comprehensive evaluation and analysis found that the model rats of chronic salpingitis with qi stagnation and blood stasis basically conformed to the change characteristics of blood stasis syndrome and the pathological manifestations of chronic inflammation, which were similar to the clinical manifestations of the disease in humans, and the modeling method was feasible.
