Objective: To investigate the effect of geniposide on the expression of IL-6, TNF-α and IL-1β in mammary tissue and mammary epithelial cells of mastitis mice. Methods Forty postpartum female mice were randomly divided into blank control group and model group, L group, M group and H group. 12h after modeling, group L was treated with low-dose geniposide, group M was treated with medium-dose geniposide, group H was treated with high-dose geniposide, blank control group and The model group was given the same amount of phosphate buffered saline solution. The intervention effect was observed 24 hours after modeling.
Results: The mRNA levels of TNF-α, IL-1β and IL-6 in the model group, L group, M group and H group were significantly higher than those in the blank control group (P<0.05). The levels of -1β and IL-6 mRNA were significantly lower than those in the model group (P<0.05); the levels of TNFα, IL-1β, and IL-6 mRNA in the M group and H group were significantly lower than those in the L group (P<0.05); The mRNA levels of IL-1β and IL-6 were significantly lower than those in the M and L groups (P < 0.05). The L, M and H groups had significantly lower TNF-α, IL-1β, and IL-6 protein levels than the model group (P < 0.05). < 0.05); the protein levels of TNF-α, IL-1β and IL-6 in group M and H were significantly lower than those in group L (P < 0.05); the protein levels of TNF-α, IL-1β and IL-6 in group H were significantly lower in group M and group L (P<0.05).
Conclusion: Geniposide can inhibit lipopolysaccharide-induced mammary gland inflammatory injury, and can reduce the levels of TNF-α, IL-1β and IL-6 inflammatory factors.
