【Animal modeling】-improvement and evaluation of rat myocardial ischemia-reperfusion injury model

Objective: To improve the preparation method of the rat model of acute myocardial ischemia-reperfusion injury, and make a comprehensive evaluation, so as to lay a good model foundation for the follow-up research on MI/RI.

Methods: Thirty-six clean-grade Wistar rats were randomly divided into 3 groups: normal group (Normal group), without intervention of left anterior descending coronary artery (LAD); sham operation group (Sham group), only threading without ligation of LAD; In the ischemia-reperfusion group (MI/RI group), the LAD was ligated for 30 min and reperfused for 120 min. After the rats were anesthetized, the electrocardiogram was recorded continuously, the tracheotomy was performed for ventilator-assisted ventilation, and the LAD of the rats was intervened by thoracotomy. comprehensive evaluation of the model.

Results: The electrocardiogram of MI/RI group had obvious ST-T dynamic changes; the level of creatine kinase isoenzyme (CK-MB) in MI/RI group increased significantly after 2 hours; compared with Sham group, the infarct size of MI/RI group The myocardial cells in the MI/RI group had obvious degeneration and necrosis, and the inflammatory cells infiltrated extensively.

Conclusion: The improved modeling method not only reduces the difficulty of surgery, but also successfully establishes a rat MI/RI model, which lays a good model foundation for subsequent MI/RI research.
