【Animal Modeling】-Construction of C57bl/6j Mouse Melanoma Lung Metastasis Model

  OBJECTIVE: To investigate the influencing factors of establishing a C57BL/6J mouse melanoma model with lung metastasis, including tumor inoculation method, cell inoculation number and tumorigenic cycle.

  Methods: The mouse melanoma cells B16F10 were cultured in vitro; 1) 18 male mice aged 6-8 weeks were selected and randomly divided into three groups, 6 mice in each group, respectively by tail vein injection, intraperitoneal injection and subcutaneous injection. Each mouse was injected with 100 μL (3×106 cells) of B16F10 cell suspension. After 2 weeks, the mice were dissected to observe the growth and metastasis of melanoma; 2) Divided into 3 groups, same as above, injected 3×106 via tail vein respectively cells, 1×106 cells, 3×105 cells, after 2 weeks, the mice were dissected and the growth and metastasis of melanoma were observed; 3) Divided into 3 groups, same as above, 1×106 cells were injected into the tail vein, respectively. The mice were dissected at 1 week, 2 weeks and 3 weeks to observe the growth and metastasis of melanoma.

  Results: 1) The success rate of lung metastasis in mice was 100% after tail vein injection of mouse melanoma cells, while no lung metastasis occurred by intraperitoneal and subcutaneous injection; 2) When the number of inoculated mice melanoma cells was 1 × 106 , the number of melanoma cells with pulmonary metastasis was moderate; when the number of inoculated cells was 3×106, the number of melanoma cells with pulmonary metastasis was too much; when the number of inoculated cells was 3×105, the number of melanoma cells with pulmonary metastasis was 3) After 1×106 mouse melanoma cells were injected into the tail vein, after 2 weeks of feeding, obvious pulmonary metastasis of melanoma cells could be observed, and the mice did not die; after 3 weeks of feeding, melanoma cells The number of cells metastasized to the lungs was too large, and more than half of the mice died; after 1 week of feeding, the number of lung metastases of melanoma cells was low.

  Conclusion: 1×106 mouse melanoma cells were injected via tail vein and grown for 2 weeks, which is the recommended method for the establishment of C57BL/6J mouse melanoma lung metastasis model.
