  Objective To study the effect of Siwu Mixture on the mouse model of primary dysmenorrhea, and to explore its possible mechanism.

  Methods Sixty healthy SPF KM female mice were randomly divided into 4 groups, namely normal group, model group, Tongjingbao granule group and Siwu mixture group. A mouse model of primary dysmenorrhea induced by estrogen combined with oxytocin was used. After 3 days of adaptive feeding, the mice were subcutaneously injected with 2 mg/kg of estradiol benzoate for 10 days. From the 4th day, the normal group and the model group were given equal doses of distilled water, the positive drug Tongjingbao granules group was given 4 g/kg, and the Siwu mixture group was given 8 mL/kg, for 7 days. Taking the number of writhing and writhing latency within 30 minutes after injection of oxytocin as the main observation indicators, the analgesic effect of Siwu Mixture was investigated, and the superoxide dismutase (SOD) and malondialdehyde (MDA) in serum were determined; Enzyme-linked immunosorbent assay (ELISA) was used to detect the contents of prostaglandin F2α (PGF2α ), interleukin 6 (IL-6) and tumor necrosis factor α (TNF-α) in uterine tissue; uterine pathological sections were prepared to observe glandular hyperplasia and inflammatory cells Infiltration, immunohistochemical detection of cyclooxygenase 2 (COX-2) expression.

  Results Compared with the normal group, the number of writhing within 30 min in the model group was significantly increased, the writhing latency was significantly shortened (P<0.01), the serum MDA level was significantly increased (P<0.01), the levels of PGF2α and TNF-α in uterine tissue were significantly increased (P<0.01). The level of IL-6 increased significantly (P < 0. 01), and the content of IL-6 increased (P < 0. 05). Endometrial glands expanded and showed papillary hyperplasia, interstitial edema, inflammatory cell hyperplasia and infiltration, and COX-2 expression. enhanced. Compared with the model group, the number of writhing within 30 min in the positive drug group and the Siwu mixture group decreased (P<0.05, P<0.01), and the writhing latency in the Siwu mixture group increased (P<0.05) , the serum MDA level in the positive drug group and Siwu mixture group decreased (P<0.01), the level of PGF2α in uterine tissue decreased (P<0.01, P<0.05), and the levels of IL-6 and TNF-α decreased (P<0.01, P<0.05).   Conclusion Siwu Mixture can significantly reduce the number of writhing in 30 min and prolong the writhing latency in mice with primary dysmenorrhea. Its mechanism may be related to the reduction of serum MDA content, PGF2α, IL-6, TNF-α levels and COX-2 levels in uterine tissue. expression and improve histopathological changes, which deserves further in-depth study.
