【Animal modeling】-The improvement effect of Baixiangdan capsule on premenstrual anxiety disorder caused by ovariectomy in rats

  Objective To compare traditional Chinese medicine Baixiangdan Capsules (BXD), serotonin 3 receptor (5-HT3R) specific agonists and their antagonists, cannabinoid receptor 1 (CB1R) specific agonists and their antagonists through a series of behavioral tests. Interventional effects of its antagonists on premenstrual dysphoric disorder (PMDD) anxiety-induced rats.

  Methods An animal model of PMDD anxiety disorder was established in rats by ovariectomy, hormone-induced estrous cycle and housing invasion. Follow-up experiments were carried out in two batches: the first batch of rats was divided into control group, model group, 5-HT3R agonist (1-phenylbiguanide, PBG)) group, CBIR agonist (WIN55212-2, WIN) group, PBG+WIN The second batch of rats were divided into control group, model group, 5-HT3R antagonist (Granisetron, GRA) group, CB1R antagonist (Rimonabant, RIM) group, GRA+RIM group, BXD group. After the drug intervention, the anxiety-like behaviors of the rats in each group were evaluated by the open field behavior test (OFT) and the elevated plus maze behavior test (EPM).

  Results Compared with the control group, the scores of residential intrusion (RIT) mixed aggression in the model group were significantly increased (P<0.01), the number of entry times and dwell time in the central area of the OFT were significantly decreased (P<0.05), and the EPM open arm entry was significantly reduced (P<0.05). The number of times and the percentage of staying time were significantly reduced (P<0.05); compared with the model group, the RIT mixed aggressive behavior scores of the PBG group, RIM group, and BXD group were significantly reduced (P<0.01), and the number of entering the central area of OFT and the staying time were significantly lower Increased (P<0.05), EPM open arm entry times and the percentage of residence time were significantly increased (P<0.05).

  Conclusion Traditional Chinese medicine BXD, 5-HT3R specific agonist 1-phenylbiguanide, and CB1R specific antagonist Rimonabant can effectively correct anxiety-like behaviors in PMDD anxiety disorder model rats.
