Objective To observe the effect of over expression of lysosome associated membrane protein 2A in substantia nigra on motor behavior of experimental monkeys induced by chaperone mediated autophagy in a Parkinson's disease rhesus monkey model.
Methods Seven rhesus monkeys were injected with Lewy bodies (LBs) extract from the brain tissue of PD patients after death (voluntary donation) into the right striatum region of the brain with stereotactic injection apparatus, and the brain tissue of non PD patients (voluntary donation) extract from the brain tissue of non PD patients after death (voluntary donation) was injected into the left striatum region to construct PD rhesus monkey models. The model animals were randomly divided into two groups: LAMP2A overexpression group (4 animals) was injected with LAMP2A overexpression adeno-associated virus vector into the substantia nigra bilaterally; The control group (3 rats) was injected with the control virus vector bilaterally in the substantia nigra area. The motor ability of PD rhesus monkey model and the effect of LAMP2A on the motor function of the PD model were evaluated by step test and plate feeding test. Determination of total cerebrospinal fluid and serum in experimental monkeys by ELISA α- Synuclein( α- synuclein, α- Syn) and phosphorylation α- Syn level was measured.
Results The fine motor level of the left upper limb in the control group was lower than that in the right side in the ladder experiment and plate feeding experiment; In the LAMP2A overexpression group, there was no significant difference in the fine motor level of the left and right upper limbs of rhesus monkeys; Determination of total cerebrospinal fluid and serum by ELISA 12 months after operation in the control group α- Syn level and phosphorylation α- Syn level increased significantly; CSF phosphorylation in LAMP2A overexpression group α- Syn level and serum phosphorylation α- The syn level was lower than that of the control group after 8 months.
Conclusion Overexpression of LAMP2A may increase α- The CMA clearance of syn improved the fine motor ability of PD rhesus monkey model.
