Objective To study the effects of different doses of isoproterenol on cardiac function and pathomorphology in rats.
Methods SD rats were randomly divided into four groups: control group, low dose model group (25 mg/kg), medium dose model group (50 mg/kg) and high dose model group (100 mg/kg). In the model group, different doses of isoproterenol were injected subcutaneously at multiple points in the neck for 2 days, while in the control group, the same amount of normal saline was injected. Twenty four hours after the last administration, the QRS interval, P wave duration and amplitude were detected by electrocardiography (ECG); TTC staining was used to detect myocardial infarction area, and HE staining was used to observe myocardial pathological morphology.
Results Compared with the normal control group, the heart weight and left ventricular weight of rats in different dose model groups were significantly increased (P<0.01); ECG showed that the duration of P wave in the high-dose model group increased (P<0.01), and the amplitude of P wave in the low-dose model group and high-dose model group increased (P<0.05, P<0.01). At the same time, there were ECG manifestations of arrhythmia. Both TTC staining and HE staining showed pathological changes of myocardial injury.
Conclusion The myocardial tissue of rats in the model group has different degrees of injury, and the myocardial injury in the high-dose model group is the most serious, which provides a basis for the preparation of animal models of ischemia/infarction like heart disease and the study of related mechanisms.
