[Animal modeling] - Application analysis of 274 rheumatoid arthritis animal models based on data mining

  Objective To study the modeling factors of rheumatoid arthritis animal models, and provide methodological reference for improving the success rate of modeling and evaluating the effectiveness of tested drugs.

  Methods With rheumatoid arthritis and animal models as the subject words, search the Chinese Journal Full Text Database (CNKI) and Wanfang Database (January 2017 to June 2020) for relevant journal literature, collect the types of experimental animals, excitation methods, inflammatory methods, test indicators, etc., establish a data base, and use Excel 2013 and SPSS Statistics 19.0 statistical software to conduct association rule analysis and factor analysis on the imported test indicators.

  Results 274 journal articles were included, among which male SD rats (69 times, 25.18%) and male Wistar rats (54 times, 19.71%) were the most frequently used experimental animals; The most commonly used inflammatory methods were CIA (172 times, 62.77%) and AA (82 times, 29.93%); The most frequently used stimulation methods were subcutaneous injection at the tail root (71 times, 25.91%) and intradermal injection at the right rear foot (52 times, 18.98%); The most frequently detected indicators were plantar swelling (133 times, 17.48%), arthritis score index (84 times, 11.04%), and TNF in serum- α Level (74 times, 9.72%) and joint histopathology (66 times, 8.67%).

  Conclusion It is suggested that male SD rats or male Wistar rats should be used in the establishment of rheumatoid arthritis animal models, and the method of subcutaneous injection at the tail root should be used to cause inflammation. The success rate of the model can be improved by using 0.1 mL of bovine type II collagen mixed with Freund's incomplete or complete adjuvant in a 1:1 emulsion. After consulting the literatures in the past three years, 274 literatures meeting the requirements were selected. This paper analyzes the existing animal models, and puts forward suggestions for further improvement of animal models of rheumatoid arthritis and standardization of model evaluation Improve the coincidence between animal model and clinical practice. Summarize the research progress of animal models of rheumatoid arthritis from the aspects of model animal selection, model type of rheumatoid arthritis, drug modeling and detection indicators, and promote the in-depth study of rheumatoid arthritis.
