Objective To establish a stable and efficient rat model of hyperuricemia with liver and kidney injury, gouty arthritis and other complications.
Methods Forty eight male Sprague Dawley rats (hereinafter referred to as SD rats) were randomly divided into 6 groups (8 rats in each group), which were blank control group (Control group), potassium oxazinate group (OA group), potassium oxazinate+hypoxanthine group (OA+H group), potassium oxazinate+yeast extract group (OA+YE group), 10% fructose+0.2% ethambutol group (10% Fru+0.2% EMB group), 2% potassium oxazinate combined with 12% yeast special feed group (OA PO group), After 5 weeks, the serum uric acid, creatinine, urea and other biochemical indicators of rats were measured, and pathological sections of kidney and joint synovium muscle were observed.
Results Compared with the blank control group, the serum uric acid level in OA PO group increased, accompanied by severe liver and kidney injury and arthritis. OA, OA+H, OA+YE, 10% Fru+0.2% EMB groups did not reach the level of high uric acid, but a certain degree of liver and kidney damage and arthritis occurred.
Conclusion 2% potassium oxazinate and 12% yeast extract mixed feed is the best way to establish hyperuricemia and its complications model. This model is also suitable for the study of hyperuricemia induced kidney damage and joint inflammation.
