【Animal Experiment】-Mice experiment proved for the first time that running helps fight cancer

Objective: To investigate the effect of low molecular weight heparin sodium (LMWH-Na) adjuvant on the humoral immune response of hepatitis A virus (HAV) vaccine in mice.

Methods: ICR mice were randomly divided into 8 groups, each with 6 mice, 5 strains of hepatitis A live attenuated vaccine HepA-1 (18EU) and 5 different doses (100μL, 20μL, 10μL, 1μL, 0.1μL)) low molecular weight Heparin sodium was used as the experimental group, physiological saline was used as the negative control group (blank), HepA-1 (18EU) was used as the positive control group, and HepA-1 mixed with Al(OH)3 (300μg) was used as the adjuvant control group for aluminum immunized mice once At 4, 8, 12, and 16 weeks after immunization, blood was collected from the tail vein, and the anti-HAV antigen-specific IgG antibody in mouse serum was detected by ELISA. During the experiment, observe the health of the mice, and after 16 weeks of immunization, collect 100 μL of the main organs and major organs of the maximum dose of low molecular weight heparin sodium in the kidney, spleen, liver, lung and heart groups and the physiological saline group. Normal saline group.

Result: Except for the negative control group, mice in each group produced anti-HAV IgG 4 weeks after immunization, and the antibody level gradually increased with time, and reached the highest value at 8 weeks and gradually decreased. The antibody levels in different groups are different at the same time. Among them, the fourth and eighth weeks were the aluminum adjuvant control group, the low-molecular-weight heparin sodium 100μL group, the low-molecular-weight heparin sodium 20μL group, and the low-molecular-weight heparin sodium 10μL group. The antibody level was higher than that of the positive control group. . (P<0.05), the 12th week, aluminum adjuvant control group, low molecular weight heparin sodium 20μL group, low molecular weight heparin sodium 10μL group, positive control group (P<0.05), 16th week, aluminum adjuvant The antibody levels of the control group and the low molecular weight heparin 10μL group were higher than the positive control group (P<0.05), but the low molecular weight heparin sodium group had higher antibody levels throughout the experiment. Lower than the aluminum adjuvant control group. Low molecular weight heparin sodium has been shown to enhance the HAV antigen-specific humoral immune response and has an adjuvant effect. During the experiment, mice in each group showed no abnormalities. No pathological changes were observed in kidney, spleen, liver, lung or heart tissue of mice in the low molecular weight heparin sodium 100μL group.

Conclusion: Low molecular weight heparin sodium can significantly enhance the humoral immune response of mouse HAV antigen, and has the value as a new vaccine adjuvant.
