[Animal modeling] Expression mode of TETs protein in urethane induced lung cancer model of mice

  Objective: To investigate the expression pattern of TET family proteins in mouse lung cancer model.

  Methods: The mice were divided into 3 groups with 10 mice in each group. They were set as control group (Con), urethane induced group (Ure), and hydroxyurea administered group (Ure HU). Ure group and Ure HU group were given 800mg/kg urethane solution for 10 consecutive weeks to induce the establishment of mouse lung cancer model. After 10 weeks, the Ure HU group was given HU drug (500mg/kg) for 21 days. Observe the number of pulmonary nodules. The expression patterns of TETs in three groups of mice were analyzed by qPCR and Western blot.

  Results: Compared with Con group, the weight of Ure and Ure HU groups decreased significantly. Morphological observation showed that both Ure group and Ure HU group had lung tissue lesions. The results of qPCR showed that the expression of TETs in Ure group decreased significantly (P<0.05). After HU treatment, the expression of TET1 and TET2 increased in Ure HU group (P<0.05). The results of Western blot and qPCR were consistent.

  Conclusion: The abnormal expression pattern of TETs protein in the murine lung cancer model induced by urethane proves that TET1 and TET2 play an important role in the development of lung cancer.
