Objective: To provide experimental methods and technical support for the establishment of the infection model of human coxsackie virus B2 in rhesus monkeys, and lay a foundation for the follow-up research on its infection mechanism, pathophysiology, etc., through the preliminary analysis and study of the infection characteristics of rhesus monkeys infected by human coxsackie virus B2 through oral cavity.
Methods: The infant rhesus monkeys aged 3-4 months were infected with Coxsackie B2 virus through oral cavity, and the clinical symptoms and temperature of rhesus monkeys were observed and recorded; Collect blood to detect physiological and biochemical indexes; Blood samples and herpes virus detection; Herpes tissues were examined by pathology.
Results: Herpes of hands, feet and mouth appeared 2~5 days after infection in rhesus monkeys; The animals are depressed, and their activities and diet are reduced; The body temperature presents the curve of heat retention; Coxsackie B2 virus can be detected in herpes fluid and blood samples; Blood routine examination showed leukopenia, increase of monocytes, decrease of lymphocytes, increase of granulocytes, increase of eosinophils and decrease of basophils. Blood biochemical tests showed that liver function, kidney function and myocardial enzymes had increased to varying degrees; The histopathology of herpes showed that the herpes of rhesus monkey showed squamous epithelial thickening, excessive surface keratosis, local necrosis with inflammatory cell infiltration, and abscess formation.
Conclusion: A series of characteristic hand foot mouth herpes, clinical symptoms, physiology and biochemistry, viremia and other conditions occur after human coxsackie B2 virus infection in infant monkeys, which indicates that rhesus monkey infant monkeys can successfully construct an animal model of coxsackie B2 virus infection through oral infection, and the relevant detection methods and technologies established are feasible, which will provide a basis for the subsequent establishment of rhesus monkey coxsackie B2 virus infection model, pathogenesis research The evaluation of drugs and vaccines laid the experimental foundation.
