[Animal modeling] - Establishment of BALB/c nude mice myeloid leukemia model by tail vein injection of M1 cells

  Objective: To construct M1 leukemia model with BALB/c nude mice

  Methods: Twelve female BALB/cNude mice aged 5-6 weeks were randomly divided into low, medium, high dose groups and blank groups, with 3 mice in each group, and injected into tail vein × 106, 5 for each × 106 and 8 each × 106 mice. Observe the general condition of mice, collect peripheral blood on the 0, 10, 20 and 30 days after modeling, test the blood routine, classify white blood cells, kill the mice on the 30 days or at the point of death, detect the proportion of CD33CD117 positive cells in peripheral blood and bone marrow by flow cytometry, and prepare histopathological sections

  Results: The mice in the model group showed lethargy, less movement and arched back on the 10th to 15th days after inoculation. On the 30th day of the experiment, compared with the blank group, the number of white blood cells in the peripheral blood of the mice in the high-dose group increased significantly (P<0.05); The proportion of peripheral blood leukemia cells in each group was significantly higher than that in the blank group (P<0.05); The proportion of CD33+CD117+in bone marrow of mice in each group increased, and that in high-dose group was the most significant (P<0.05); A small amount of leukemic cells can be seen in the spleen of the high-dose group

  Conclusion: BALB/cNude mice were injected via tail vein 8 × The acute myeloid leukemia model can be built after 106 mouse leukemia cells M1, which conforms to the biological characteristics of acute myeloid leukemia
