[Animal modeling] - Expression and significance of thrombomodulin in deep vein thrombosis model of rats

  Objective: To observe the expression and significance of thrombomodulin (TM) in the natural evolution of deep vein thrombosis (DVT) in rats

  Methods: SD rats were randomly divided into the model group (n=60) and the control group (n=6). The model group used the "stenosis method" to establish the DVT model, and the control group used the "sham operation method". After modeling, the rats were killed at 10 time points: 1h, 4h, 6h, 12h, 24h, 3d, 7d, 14d, 21d, 28d. The ratio of thrombus weight to length, the concentration of soluble TM in plasma, the expression of TMmRNA in thrombus tissue and inferior vena cava endothelium were detected

  Results: 1) The ratio of thrombus weight/length was low at the early stage of modeling (1, 4, 6, 12 h), and there was no significant difference among the four groups (P>0.05); After 24 hours, 3 days and 7 days of modeling, it was stable at a higher level (P>0.05); 14 days after modeling, it began to decrease, but there was no significant difference with the results of 21 and 28 days (P>0.05); In 24h, 3d and 7d, it was lower than that in the control group (P<0.01); 21="" 28="" at="" and="" there="" was="" no="" significant="" difference="" compared="" with="" the="" control="" group="" p="">0.05). (4) Thrombus weight/length ratio was negatively correlated with endothelial TMmRNA (r=-0.92, P<0.01); There was a positive correlation between thrombotic TMmRNA and plasma soluble TM (r=0.96, P<0.01)

  Conclusion: The change of endothelial TMmRNA expression can reflect the progress of DVT and has a good prognostic effect
