[Animal modeling] - Comparison of bronchial asthma models induced by different doses of ovalbumin in BALB/c mice

Objective: To investigate the effects of different doses of ovalbumin on serum IgE, IL-4 and IFN in BALB/c mice with bronchial asthma γ The content, the number of eosinophils in bronchoalveolar lavage fluid and the influence of lung histopathology, initially determine a more appropriate OVA modeling dose

Methods: BALB/c mice were randomly divided into normal group and model group 1 (OVA20 μ g) , model group 2 (OVA50 μ g) , model group 3 (OVA100 μ g) , dexamethasone acetate group (OVA100 μ g) In addition to the normal group, other groups of mice were sensitized by intraperitoneal injection of corresponding dose of OVA, and challenged with 1% OVA aerosol to establish a bronchial asthma model. Serum IgE, IL-4, IFN were detected by ELISA- γ The number of EOS in BALF was calculated by Rexhlet staining, and the pathological changes of lung tissue were observed by HE staining

Results: Compared with the normal group, serum IgE, IL-4, IFN of mice in each group- γ And BALF, EOS increased significantly (P<0.05), inflammatory cell infiltration, bronchial epithelial cell degeneration, bronchial smooth muscle cell proliferation and other pathological changes appeared in lung tissue- γ In terms of content and pathological changes of lung tissue, OVA50 μ Group g mice are more obvious

Conclusion: All three doses of OVA can successfully induce asthma in BALB/c mice, 50 μ The asthma model induced by gOVA is more obvious, and it is recommended to be the appropriate modeling dose for BALB/c mouse asthma model
