Objective: To investigate the method of establishing quail gout model by simulating clinical high protein and high calcium diet to induce Difak quail gout, so as to provide basis for the study of gout pathogenesis and screening of prevention and treatment drugs
Methods: Thirty six 30 day old male healthy Difak quails were randomly divided into three groups according to body weight and blood uric acid level, namely, control group, model I group and model II group, with 12 quails in each group. The control group quails were fed with basic diet, the model group was fed with high protein and high calcium diet (diet crude protein content 29.5%, calcium content 7.8%), the control group and model I group drank freely, the model II group was restricted to drinking water for 4 hours a day, and the experiment lasted for 50 days, At the same time, renal function was detected and histological pathological sections were observed
Results: High protein and high calcium diet could significantly increase the serum uric acid level of quails; Uric acid in feces and urine decreased significantly; Serum creatinine and urea nitrogen levels increased; It leads to obvious deposition of uric acid salt in quail kidney. High protein and high calcium diet combined with restriction of drinking water can aggravate the increase of blood uric acid level and deposition of renal uric acid salt, causing some quails to have gout symptoms such as swollen joints and deformation
Conclusion: The gout quail model can be established by feeding high protein and high calcium diet and limiting the amount of drinking water, and its pathogenesis is more consistent with the etiology, pathogenesis and clinical manifestations of gout, providing a reference for the basic research and drug development of gout
