[Animal modeling] Study on the therapeutic effect and recurrence of GnRH-a plus estrogen on rat endometriosis model

  Objective: To investigate the therapeutic effect of GnRH-a plus different doses of estrogen on endometriosis and its influence on recurrence

  Methods: Fifty one rats with endometriosis were divided into five groups: control group (n=10), which was injected with 1 mg/kg normal saline subcutaneously once; Study group: anti dose group I (low dose estrogen group n=10), anti dose group II (medium dose estrogen group n=10), anti dose group III (high dose estrogen group n=10), GnRH-a group (n=11). The study group was given leuprorelin 1mg/kg subcutaneous injection for one time, and the anti dose groups I, II and III were given different doses of estradiol valerate (25 μ g/kg、50 μ g/kg、100 μ G/kg). At the same time, GnRH-a group was given normal saline by gavage, 3mL/animal, once a day in each study group, for a total of 14 days. At the 4th and 7th weeks after administration, open the abdomen to explore, measure the volume of endometriosis, take the tissue of endometriosis, and detect TNF in endometriosis by immunohistochemistry- α、 The expression of StAR, the blood collection method of orbital medial canthus venous plexus, and the serum CA125 level were detected by enzyme-linked immunosorbent assay

  Results: Before administration, there was no significant difference in the volume of ectopic lesions in rats of each group (P>0.05).   Conclusion: GnRH-a plus low-dose estrogen has a good effect on rat endometriosis model, and the reverse addition of medium and high-dose estrogen may induce TNF- α、 StAR promotes the recurrence of endometriosis in rats
