Objective: To explore the factors influencing the establishment of C57BL/6J mouse melanoma lung metastasis model, including the tumor inoculation method, the number of cells inoculated and the tumor forming cycle
Methods: B16F10 mouse melanoma cells were cultured in vitro; 1) 18 male mice aged 6~8 weeks were randomly divided into three groups, with 6 mice in each group. They were injected by tail vein, intraperitoneal injection and subcutaneous injection respectively. Each mouse was injected with 100 μ L(3 × 106 cells) B16F10 cell suspension. After 2 weeks, dissect the mice and observe the growth and metastasis of melanoma; 2) Divide into three groups, as above, and inject 3 through tail vein respectively × 106 cells, 1 × 106 cells, 3 × 105 cells. After 2 weeks, the mice were dissected and the growth and metastasis of melanoma were observed; 3) Divide into three groups, as above, and inject 1 through tail vein × 106 cells were dissected at 1 week, 2 weeks and 3 weeks respectively to observe the growth and metastasis of melanoma
Results: 1) The success rate of pulmonary metastasis in mice was 100% after tail vein injection of melanoma cells, while intraperitoneal injection and subcutaneous injection did not cause pulmonary metastasis; 2) The number of melanoma cells inoculated in mice is 1 × At 106, the number of melanoma cells with lung metastasis was moderate; The number of inoculated cells is 3 × At 106, the number of melanoma cells with lung metastasis was excessive; The number of inoculated cells is 3 × At 105, the number of melanoma cells with pulmonary metastasis was less; 3) Tail vein injection 1 × After feeding 106 melanoma cells in mice for 2 weeks, obvious lung metastasis of melanoma cells could be observed, and the mice would not die; After feeding for 3 weeks, the number of melanoma cells metastasized to the lungs was excessive, and more than half of the mice died; After one week of feeding, the number of pulmonary metastasis of melanoma cells was small
Conclusion: Injection through tail vein 1 × 106 mouse melanoma cells were grown for 2 weeks, which is the recommended method for constructing C57BL/6J mouse melanoma lung metastasis model
