Objective: To establish a model of spontaneously hypertensive rats (SHR) with hyperlipidemia by feeding them with high-fat diet, and to evaluate their heart and kidney injuries, so as to provide pharmacodynamic animal models and relevant methods for the evaluation of therapeutic drugs for hypertension with hyperlipidemia.
Methods: Three week old SHR patients were randomly divided into blank control group and model group; At the same time, Wistar Kyoto (WKY) rats with normal blood pressure at the same age were selected as the model control group. The blank control group and model control group were fed with normal maintenance feed, while the model group was fed with high-fat feed to induce SHR to produce hypertension and hyperlipidemia. The blood pressure and body weight of rats in each group were measured regularly during the modeling period, and the blood pressure of rats was measured at the modeling end point? After blood lipid and body weight were killed, heart and kidney tissues were collected and their fibrosis damage was detected.
Results: After 23 weeks of feeding with SHR high fat diet, the blood lipid level was obviously disturbed; The heart and kidney organs showed obvious pathological changes: obvious hypertrophy of myocardial cells, significant reconstruction of blood vessel wall of interlobular artery in renal tissue, and severe fibrosis of heart and kidney tissues.
Conclusion: SHR can successfully obtain the rat model of hypertension combined with hyperlipidemia after 23 weeks of feeding with high-fat diet. The target organs of heart and kidney show obvious pathological changes, which are similar to the clinical manifestations of human hypertension combined with hyperlipidemia. SHR is expected to be widely used as a drug to treat hypertension combined with hyperlipidemia. Or to improve the pharmacodynamic evaluation model of the therapeutic drugs for the cardiac and renal system damage of such complications.
