Objective: To establish an animal model of chemical chronic nonbacterial prostatitis, and provide a reliable model animal and evaluation method for the pathogenesis and drug research of CNP.
Methods: SD male rats were randomly divided into control group and model groups A, B and C. Model groups A, B and C were respectively injected with 1% sterile carrageenan into the left and right ventral lobes of the prostate for 20, 50 and 100 μ L; 50 for each control group μ L Sterilized normal saline solution. After 7 days of post-treatment, respectively from the anatomical point of view, prostate index, leukocyte detection, pathological section and tumor necrosis factor- α (TNF- α)、 Nuclear factor- κ B (NF- κ B)、I κ B kinase (IKK α)、 Phosphorylated IKB- α (p-IKB- α) And cyclooxygenase-2 (COX-2) protein expression.
Results: Compared with sham operation group, the softness, elasticity and adhesion of prostate tissue in model group A, B and C decreased; Prostate index and total number of white blood cells increased significantly (P<0.01). The increase rates of prostate index in group A, B and C were 21.1%, 61.7% and 72.7% respectively; The total number of white blood cells increased by 75.0%, 103.6% and 114.8%, respectively; The pathological results showed that the prostatic interstitium in group A was less swollen, the interstitium in group B and C was significantly swollen, the prostate in group C was significantly atrophied, and some glands were degenerative and necrotic; A. NF in prostate tissue of group B and C- κ B、IKK α、 p-IKB- α、 TNF- α And COX-2, NF- κ The B inflammatory pathway is activated.
Conclusion: The left and right ventral lobes of the prostate of rats were injected with 1% carrageenan 20, 50 and 100 respectively μ 50. They can induce prostatitis reaction of different degrees, but μ The L dose is too small, the inflammatory reaction is weak, and the operation error is large; one hundred μ L The inflammatory reaction is too strong, and some animals die; fifty μ The animal model of prostatitis induced by L dose is the most successful, and can obviously activate NF- κ B Inflammation signal pathway.
