Objective: To establish a collagen induced arthritis model in DBA/1J mice and identify the model from morphology, pathology and immunology.
Methods: Bovine type II collagen was mixed with adjuvant and fully emulsified, and the DBA/1J mice were immunized subcutaneously at the tail root for the first time. The same method was used to immunize again 21 days later. Observe the weight change, joint swelling degree and other morphological indicators of mice, conduct ankle joint pathological examination, and detect the level of cytokines in serum with CBA method.
Results: The weight of mice in the model group began to drop 35 days after the first immunization and was continuously lower than that of the control group. On the 28th day, the mice in the model group had red and swollen feet, and on the 35th day, the swelling thickness of feet was significantly higher than that in the control group (P<0.05), and on the 62nd day, the incidence rate was as high as 90%. Pathological examination showed that the joint cavity of the model group mice became narrow, synovial tissue proliferated, inflammatory cells infiltrated, and cartilage structure was damaged to varying degrees. Compared with the control group, the serum inflammatory factors IL-6 and IFN in the model group mice- γ、 MCP-1 increased significantly (P<0.05), while="" the="" anti-inflammatory="" factor="" il-10="" decreased="" slightly="" p="">0.05).
Conclusion: The CIA model of DBA/1J mice can be successfully induced by subcutaneous injection of bovine type II collagen into the tail root.
