[Animal modeling] - Effect of electroacupuncture on type 2 diabetes mellitus with renal hypertension in rats

  Objective: To observe the therapeutic effect of electroacupuncture on type 2 diabetes mellitus with renal hypertension in rats, explore its possible mechanism, and further improve the application of acupuncture in clinical acute and chronic diseases.

  Methods: 50 Wistar rats were randomly divided into simple diabetes group, simple renal hypertension group, compound model electroacupuncture group and compound model non electroacupuncture group. After 4 weeks of high-fat and high-sugar diet, the rats were injected with streptozotocin (STZ) rapidly intraperitoneally to make a simple type 2 diabetes model. After the blood glucose of the model was stabilized, the improved "two kidneys and one clip method" was used to ligate the unilateral renal artery of the rats, causing renal artery stenosis and forming renal hypertension. Finally, a stable compound model of type 2 diabetes mellitus with renal hypertension was obtained. Among them, except that the rats in the non electroacupuncture group of the compound model were not treated with electroacupuncture, the rats in the other groups were treated with electroacupuncture for two cycles (2 weeks). Finally, the changes of blood pressure (BP), fasting blood glucose (FBG), blood creatinine (Cr), urea nitrogen (BUN), glycosylated hemoglobin (GHbA1c), renin (PRA) and angiotensin Ⅱ (Ang Ⅱ) values of rats in each group before and after electroacupuncture were detected.

  Results: After 2 weeks of electroacupuncture, the values of BP, FBG, GHbA1c, PRA, Ang Ⅱ in the electroacupuncture group of the composite model were lower than those in the non electroacupuncture group of the composite model (P<0.01), but="" the="" values="" of="" cr="" and="" bun="" were="" not="" significantly="" changed="" p="">0.05).

  Conclusion: Electroacupuncture stimulation of Zusanli points on both sides of the model rats with type 2 diabetes mellitus and renal hypertension can reduce the blood sugar and blood pressure of the compound model rats to normal levels, and within 2-3 days after a single electroacupuncture stimulation, the blood sugar and blood pressure of the compound model rats can be stabilized at the level after treatment. In addition, acupuncture and moxibustion therapy has the advantages of economy, safety and no side effects compared with other treatment methods, and is more suitable for the early clinical treatment and basic research of type 2 diabetes mellitus with renal hypertension.
