[Animal modeling] - Comparative study of spontaneous and induced type 2 diabetes mouse models

  Objective: To compare the differences in glucose metabolism, insulin resistance and inflammatory reaction between spontaneous and induced type 2 diabetes mouse models.

  Methods: The animals were divided into normal control group, induced model group and spontaneous model group with 10 animals in each group. Normal control group: normal 11 week old C57BL/6J mice; Induced model group: 11 week old C57BL/6J mice were induced with high-fat diet combined with STZ to establish diabetes model; Spontaneous model group: C57BL/KsJ db/db mice aged 8 weeks were selected; The changes of body weight, fasting blood glucose and glucose tolerance were monitored within eight weeks; FINS, AUC, HOMA-IR and TNF before and after the test- α, IL-18,IL-1 β, INF- γ Change of content; Eight weeks later, the animals were killed to take liver, kidney, pancreas, testis, skeletal muscle and other tissues for pathological analysis.

  Results: During the experimental period, the body weight of the spontaneous model group continued to rise, while that of the induced model group decreased significantly; The blood glucose of mice in the induced model group decreased after eight weeks, while the mice in the spontaneous model group maintained high blood glucose steadily; FINS in induced model group increased, AUC and HOMA-IR decreased significantly, FINS in spontaneous model group did not change significantly, AUC and HOMA-IR increased significantly; At the beginning of the establishment of the model, the serum proinflammatory factor (TNF) in the two models- α, IL-18,IL-1 β, INF- γ) The content of β - lactamase was higher than that of normal control group. At the eighth week, the content of proinflammatory factors in the induced model group decreased, while the content of proinflammatory factors in the spontaneous model group increased significantly; The pancreas and other tissues of the two models showed obvious inflammatory lesions.

  Conclusion: Eight weeks later, the abnormal glucose metabolism and inflammatory reaction in the spontaneous type 2 diabetes model are getting worse, and hyperglycemia symptoms are maintained for a long time; After eight weeks of feeding, the induced type 2 diabetes model partially alleviated glucose metabolism and inflammatory reaction, and partially improved hyperglycemia symptoms.
