[Animal modeling] - Establishment of orthotopic liver cancer transplantation model in nude mice

  Objective: To establish an orthotopic liver cancer transplantation model in BALB/c nude mice.

  Methods: Experimental animals were anesthetized with isoflurane gasification, and BALB/c nude mice models of orthotopic liver cancer transplantation were established by "embedding method" and "sandwich method" respectively. After operation, the growth of tumor tissue was detected with small animal living imaging system, and the liver and tumor tissue were histopathologically examined 4 weeks later.

  Results: The BALB/c nude mouse model of orthotopic liver cancer transplantation established by sandwich method was characterized by short operation time, low difficulty, high tumor formation rate, good growth of tumor tissue, no extensive tumor planting in the abdominal cavity, no abnormality in histopathological examination, and low animal mortality.

  Conclusion: Compared with the embedding method, the sandwich method has more advantages. It can rapidly prepare a large number of orthotopic liver cancer transplantation models in mice, providing a more convenient platform for the orthotopic pharmacodynamics of liver cancer and other research.
