[Animal modeling] - Comparative study on three severe pancreatitis models of rats with different severity

  Objective: To provide the basis for the researchers to select the appropriate concentration of sodium taurocholate to prepare SAP model of severe pancreatitis in rats.

  Methods: Sixty SD rats were randomly divided into sham operation group, 1.5% concentration group, 3.5% concentration group and 5% concentration group. SAP models were prepared by retrograde biliopancreatic duct injection of 1.5%, 3.5% and 5% sodium taurocholate respectively in each group. The mortality of rats in each group was calculated after operation; Detection of serum amylase and tumor necrosis factor- α (TNF- α)、 Interleukin-6 (IL-6) level; Observe the pathological score of HE staining in pancreatic tissue of rats in each group.

  Results: The mortality of the 5% concentration group was significantly higher than that of the 1.5% concentration group- α (TNF- α)、 The levels of interleukin-6 (IL-6), the pathological scores of hemorrhage and acinar necrosis were significantly higher than those of 1.5% and 3.5% concentration groups.

  Conclusion: 5% sodium taurocholate retrograde biliopancreatic duct injection can better prepare SAP model, and more in line with the physiological and pathological manifestations of SAP.
