[Animal modeling] - Establishment and evaluation of rat model of primary immune thrombocytopenic purpura with excessive heat

  Objective: To establish a rat model of peripheral blood thrombocytopenia in accordance with the "heat excess syndrome" of traditional Chinese medicine and primary immune thrombocytopenic purpura.

  Methods: SD rats were injected with 20% dry yeast suspension at multiple points on the back and 1:4 diluted rabbit anti SD rat platelet serum (APS) intraperitoneally to establish the model of primary immune thrombocytopenic purpura fever in rats. The characteristics of TCM syndromes, hemogram, bone marrow picture and the level of 5-hydroxytryptamine (5-HT) in the thermoregulatory center of thalamus were observed.

  Results: In the model group, the body temperature and daily water consumption increased significantly 2 to 6 hours after injection, and purpura of toes appeared on the 4th day of modeling and intestinal mucosal hemorrhage appeared on the 30th day of modeling (P<0.05); The number of platelets in peripheral blood cells and megakaryocytes in bone marrow cells decreased significantly (P<0.05); The level of 5-HT in the brain thermoregulatory center increased significantly (P<0.05).

  Conclusion: The model of combination of disease and syndrome in rats with primary immune thrombocytopenic purpura and heat hyperactivity basically reflects the pathological characteristics of purpura in rats with primary immune thrombocytopenic purpura caused by heat hyperactivity.
