Objective To study the effects of shionone on the expression of Toll like receptor and immune response in ovalbumin induced asthmatic SD rats.
Methods 24 young SD rats were randomly divided into control group, model group, shionone group and dexamethasone group with 6 rats in each group. The behavioral changes of young rats were observed. The pathological changes of lung tissues after treatment were observed by hematoxylin eosin (HE) staining. The expression of TLRs mRNA in plasma was detected by qPCR; Detection of inflammatory cytokine TNF in serum by ELISA- α、 IL-1 β、 The expression level of IL-6, IL-4 and IL-13.
Results The young rats in the model group had shortness of breath, purple nose and mouth when sneezing, secretion of mucus, and irritability; The bronchial mucosa is congestive and edematous, the bronchial epithelial cells fall off, and a large number of inflammatory cells infiltrate around the airway and lung tissue. After treatment with shionone and dexamethasone, the young rats breathed more slowly, the secretion of mucus was reduced, and the hyperactivity and irritability were also reduced, but the aggression of the dexamethasone group was enhanced; The degree of congestion and edema of bronchial mucosa was relieved, and the number of inflammatory cells around the airway and lung tissue was also reduced. Compared with the model group, the expression of TLR1, TLR9, TLR10 mRNA in shionone group and dexamethasone group was significantly increased (P<0.01), and the expression of TLR2, TLR3, TLR4, TLR5, TLR6, TLR7, TLR8 mRNA was significantly decreased (P<0.01), TNF- α、 IL-1 β、 The levels of IL-6, IL-4 and IL-13 decreased significantly (P<0.01).
Conclusion Asterone can improve the lung inflammation of OVA induced asthmatic SD young rats, and change the expression of TLRs, which lays a foundation for the follow-up study of the mechanism of Asterone on TLRs signal pathway.
