[Animal modeling] - Study on the mechanism of Herba Siegesbeckiae Tongshuan Capsule in alleviating cognitive function damage in rats with chronic cerebral ischemia

  Objective To study the effect of Siegesbeck Tongshuan capsule on the rat model of chronic cerebral ischemia and blood brain barrier.

  Methods SD rats were used to make chronic cerebral ischemia model by ligating bilateral common carotid arteries. Water maze was used to evaluate the learning and cognitive function of rats, Nissl staining was used to observe the neurons in hippocampus, immunofluorescence staining was used to observe the damage degree of blood-brain barrier, and ELISA and Western blot were used to detect the content of blood-brain barrier related proteins in plasma and brain tissue respectively.

  Results Siegesbeck Tongshuan capsule could improve the decline of learning and cognitive function of rats after chronic cerebral ischemia, reduce the decrease of Nissl bodies and IgG leakage in neurons of hippocampus, reduce the levels of MMP-9, claudin-5 and occludin in plasma, and increase the expression of claudin-5 and occludin in brain tissue.

  Conclusion Siegesbeck Tongshuan capsule can reduce the damage of blood brain barrier and improve the learning and cognitive function after chronic cerebral ischemia.
