Purpose based on TLRs/NF- κ B signal pathway was used to explore the effect of Bushen Yiqi Huayu Granule on angiogenesis in osteoporosis rats.
Methods 60 SPF female SD rats were randomly divided into sham operation group, model group, granule group and positive control group, with 15 rats in each group. Except the sham operation group, the other three groups of rats were subjected to bilateral ovariectomy to prepare osteoporosis model. The sham operation group was only operated, and the control group was not ovariectomized. The rats in the granule group were given 5 g/kg Bushen Yiqi Huayu Granule by gavage, the rats in the positive control group were given 0.5 mg/kg alendronate by gavage, and the rats in the model group and the sham operation group were given the same amount of normal saline by gavage. After 12 weeks of continuous administration, the rats were killed. Bone mineral density, HE staining, Micro CT three-dimensional reconstruction analysis, CD31 fluorescence immunoassay and blood perfusion imaging analysis were performed on the femur tissue of the rats; Detection of VEGF and HIF-1 in serum by ELISA α Concentration; Western blot was used to detect the expression of p50/p-p50 and p65/p-p65 proteins.
Results Compared with the sham operation group, the model group had lower bone mineral density (P<0.05), sparse bone trabecular growth, slower angiogenesis, and fewer VEGF and HIF-1 in serum α The concentration of p50/p-p50 and p65/p-p65 protein in femur decreased significantly (P<0.05); Compared with the model group, the rats in the granule group and the positive control group had good growth of bone trabeculae, more angiogenesis, significantly increased bone density, VEGF and HIF-1 in serum α The level of p50/p-p50 and p65/p-p65 protein expression decreased significantly (P<0.05).
Conclusion Bushen Yiqi Huayu Granule may regulate TLRs/NF- κ B signal pathway can promote angiogenesis in osteoporosis rats and play an important role in relieving osteoporosis.
