[Animal modeling] - Effect of arginine vasopressin 1b receptor knockout on anxiety and social behavior of female rats

  Objective to investigate the effects of arginine vasopressin 1b receptor gene knockout on plasma corticosterone content, anxiety and social behavior in female rats.

  Methods The female SD rats with ARV 1b receptor knockout and wild-type SD rats were used as research objects. First, the expression of ARV 1b receptor in the paraventricular nucleus and supraoptic nucleus of WT and AvpR1bKO rats was detected by in situ hybridization; The plasma corticosterone content of WT and AvpR1bKO rats at rest was analyzed by high performance liquid chromatography tandem high resolution mass spectrometry; The anxiety level of rats was evaluated by the time and times WT and AvpR1bKO rats entered the central area in the open field experiment, and the time and times WT and AvpR1bKO rats entered the open arm in the elevated cross maze experiment; The social ability of WT and AvpR1bKO rats was evaluated by the time of exploring the strange rats in the three box experiment; The social novelty of WT and AvpR1bKO rats was evaluated according to the time of exploring unfamiliar rats with different familiarity.

  Results There was no expression of arginine vasopressin 1b receptor in the paraventricular nucleus and supraoptic nucleus of AvpR1bKO rats; In resting state, the plasma corticosterone content of AvpR1bKO rats had no difference compared with WT rats (P>0.05); In the open field experiment, there was no difference between AvpR1bKO rats and WT rats in the time and times of entering the central area (P>0.05); In the elevated experiment, the time and times of AvpR1bKO rats entering the open arm area had no difference from WT rats (P>0.05); In the three box experiment, there was no significant difference between AvpR1bKO rats' social competence index and WT rats' social competence index (P>0.05); At the same time, there was no significant difference between the social novelty index of AvpR1bKO rats and that of WT rats (P>0.05).

  Conclusion arginine vasopressin 1b receptor knockout does not affect plasma corticosterone content, anxiety level, social ability and social novelty in female rats at rest.
