[Animal modeling] - The intervention effect of flavonoids in Sandiaojin on ulcerative colitis in mice and its effect on intestinal flora

  Objective To investigate the effects of total flavones of Sandiajin and its monomers, calycosin and sessile, on ulcerative colitis (UC) induced by dextran sodium sulfate (DSS) in mice and intestinal flora.

  Methods The UC mouse model was established with 3% DSS. Thirty C57BL/6 mice were divided into normal group, model group, total flavone of three points of gold group (500 mg/kg), pistil isoflavone group (100 mg/kg) and sessile group (100 mg/kg). Observe the weight change of mice, carry out disease activity index (DAI), colon histopathology score, and detect IL-1 in mouse serum β And IL-6 content. The changes of intestinal flora in mice were analyzed by high-throughput sequencing.

  Results The total flavones, pistil isoflavones and stylosanthin could effectively reduce the symptoms of colitis and the expression of inflammatory factors in mice; The results of flora sequencing showed that the total flavones, pistil isoflavones, and strobilin significantly increased the diversity of intestinal flora. At the family level, the main performance was that the proportion of pathogenic bacteria was reduced, while the proportion of Muribagulaceae and Trichospiriceae was increased; At the genus level, the number of lactic acid bacteria increased and the number of conditional pathogenic bacteria decreased; At species level, it mainly showed the reduction of opportunistic pathogenic bacteria Bacteroides and Escherichia coli.

  Conclusion The total flavones, pistil isoflavones and stylosanthin have a certain improvement effect on the signs and inflammatory factors of UC mice, and have a good recovery effect on intestinal flora.
