Objective: To immunize mice with recombinant protein and dendritic cells (DC) and observe its protective effect on systemic Candida albicans.
Methods: Prokaryotic expression and induced expression of Candida albicans Sap2 protein; Candida albicans spores and mycelium suspensions are sensitive to mouse DCs. The mice were then divided into 5 groups and immunized with recombinant protein 3 times to reconstruct sensitized DCs , Sensitized DC, DC, Sap2, Sap2, PBS and Candida albicans systemic infection model to observe survival time, kidney pathological section, kidney-negative bacteria count, CD4 + and detection CD8 + percentage and observe its protective effect.
Result: The pET32a-Sap2 prokaryotic expression vector was successfully constructed, induced to express the target protein, and collected and purified. Recombinant protein, sensitized DC, DC and Sap2 protein effectively prolonged the survival time of mice infected with systemic Candida albicans, protected the mouse kidneys and enhanced the mice's immunity. Among them, multi-component combination immunity is more effective.
Conclusion: Multi-component vaccination can well resist systemic Candida albicans infection, and is the basis for research on new multi-component vaccines.
