Objective To investigate the neuroprotective effect of soybean saponin Bb preconditioning on cerebral ischemia/reperfusion in rats.
Methods SD rats were divided into sham operation group, model group and low and high dose SSBb pretreatment groups, with 15 rats in each group. The middle cerebral artery occlusion (MCAO) rat model was established by thread embolism method, and reperfusion was achieved 2 hours after occlusion to simulate brain I/R. Low and high dose SSBb pretreatment groups were given 20 and 50 mg/kg SSBb by gavage 1 h before MCAO. After 24 hours of reperfusion, the neurological deficit was evaluated with modified neurological severity score (mNSS), the cerebral infarction volume was measured with 2,3,5-triphenyltetrazolium chloride (TTC), the neuronal damage was evaluated with microtubule associated protein-2 (MAP-2) staining, and the brain water content was measured with dry/wet weighing, Detection of tumor necrosis factor in ischemic penumbra by immunofluorescence- α (tumor necrosis factor- α, TNF- α) Western blot was used to detect the expression levels of postsynaptic density 95 (PSD - 95), synapsin I, and synaptogmin.
Results Compared with the model group, pretreatment with low and high doses of SSBb could significantly improve the neurological deficit, relieve brain edema and cerebral infarction, and reduce TNF in ischemic penumbra of MCAO model rats- α The expression level of PSD-95, synapsin I and synaptotagmin (both P<0.01 and P<0.001); The high dose group had the most significant effect (P<0.001).
Conclusion Preconditioning with SSBb plays a neuroprotective role in brain I/R injury in rats.
