[Animal modeling] - Effect of Jiayankangtai Granule on immune related cytokines in rats with autoimmune thyroiditis

  Objective To investigate the effect of Jiayankangtai Granule on immune related cytokines in rats with autoimmune thyroiditis, and explore the mechanism of Jiayankangtai Granule in improving autoimmune thyroiditis.

  Methods 30 female Lewis rats were randomly selected from 10 normal rats, and the remaining 20 rats were injected with thyroglobulin subcutaneously and iodine water orally for 7 weeks. The successful rats were randomly divided into model group (10 rats) and drug group (10 rats). After the continuous intervention of Jiayankangtai granules for 8 weeks, the thyroid antibody level was detected by ELISA; HE staining was used to observe the pathological changes of thyroid gland; The gene expression of target factor was detected by qPCR; Antibody microarray was used to detect the expression of target proteins and draw clustering heat map.

  Results TPOAb and TGAb in the model group were significantly higher than those in the normal group, and TPOAb and TGAb in the drug administration group were significantly lower than those in the model group. The thyroid follicular cells in the model group were destroyed and a large number of lymphocytes were infiltrated. The pathological changes in the thyroid of the drug group were alleviated compared with the model group. Model group IFN- γ、 TNF- α、 The expression levels of IL-17, CCL3, CX3CL1 genes were higher than those in the normal group. IL-4, IFN in the treatment group- γ、 TNF- α、 IL - 17, CCL3 and CX3CL1 decreased compared with the model group. The cytokine clustering among groups was good. The expression of IL-17, CCL3, CX3CL1 protein in the model group was higher than that in the normal group. IL-4, IFN in the treatment group- γ、 TNF- α、 CCL3 and CX3CL1 are lower than the model group.

  Conclusion Jiayankangtai Granule can regulate immune related cytokines and alleviate thyroid damage in rats with autoimmune thyroiditis.
