Objective C57BL/6J mice were anesthetized with different doses of Zoletil combined with Cyperazine hydrochloride to observe the anesthesia related time of mice under different anesthetic doses, so as to provide reference for various mouse operations and related experimental designs.
Methods 100 C57BL/6J mice (half male and half female) were randomly divided into 5 groups, 20 mice in each group (half male and half female). The mice in the four treatment groups were intraperitoneally injected with Zoletil 55 mg/kg+Cyperazine hydrochloride 13.75 mg/kg, Zoletil 65 mg/kg+Cyperazine hydrochloride 16.25 mg/kg, Zoletil 75 mg/kg+Cyperazine hydrochloride 18.75 mg/kg, Zoletil 85 mg/kg+Cyperazine hydrochloride 21.25 mg/kg; After the righting reflex of the mice disappeared, one drop of Xinledun eye drops (about 20 µ L) was applied to each eye, and the mice were placed on the heat preservation pad. The control group mice were intraperitoneally injected with 200 µ L 0.9% sodium chloride solution. Observe, record and compare the induction time, maintenance time and recovery time of anesthesia of mice in each group. The mice were raised for 1 day after recovery from anesthesia, the serum was taken, and the liver and kidney functions were measured with Hitachi 7080 automatic biochemical analyzer: glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, creatinine.
Results The successful rate of anesthesia of C57BL/6J mice with Zoletil combined with Cyperazine hydrochloride was 100%. When the dosage of Zoletil was 55-75 mg/kg, it could quickly and safely induce anesthesia in mice, and the anesthetic effect was good. The duration of anesthesia was in direct proportion to the amount of Zoletil injection (r2=0.827 for male rats, r2=0.841 for female rats, P<0.05), and the duration of anesthesia induction was in inverse proportion to the amount of injection (r2=0.432 for male rats, r2=0.410 for female rats, P<0.05). after="" recovery="" from="" there="" was="" no="" significant="" difference="" in="" liver="" and="" kidney="" function="" indexes="" between="" each="" group="" the="" control="" p="">0.05).
Conclusion The anaesthesia of C57BL/6J mice with 55 ~ 75 mg/kg Zoletil combined with Cyperazine Hydrochloride can obtain better anaesthesia effect and safety, and the anaesthesia time can be controlled by adjusting the anesthetic dose, which suggests that this is a stable and reliable anaesthesia method.
