[Animal modeling] - The improvement effect of Baixiangdan capsule on premenstrual anxiety caused by ovariectomy in rats

  Objective To compare the intervention effects of Baixiangdan capsule (BXD), 5-HT3R specific agonist and its antagonist, and cannabinoid receptor 1 (CB1R) specific agonist and its antagonist on premenstrual dysphoria and anxiety rats through a series of behavioral tests.

  Methods The rat model of PMDD anxiety was established by ovariectomy, hormone induced estrous cycle and residential invasion. The following experiments were conducted in two batches: the first batch of rats were divided into control group, model group, 5-HT3R agonist (PBG) group, CBIR agonist (WIN55212-2, WIN) group, PBG+WIN group and BXD group; The second batch of rats were divided into control group, model group, 5-HT3R antagonist (GRA) group, CB1R antagonist (RIM) group, GRA+RIM group and BXD group. After the drug intervention, the anxiety like behaviors of rats in each group were evaluated by open field behavior test (OFT) and elevated cross maze behavior test (EPM).

  Results Compared with the control group, the mixed aggressive behavior score of residential invasion (RIT) in the model group was significantly higher (P<0.01), the number of entries into the central area of OFT and the stay time were significantly lower (P<0.05), and the number of entries into the open arm of EPM and the percentage of stay time were significantly lower (P<0.05); Compared with the model group, PBG group, RIM group and BXD group had significantly lower RIT mixed aggressive behavior scores (P<0.01), significantly higher OFT central area entry times and stay time (P<0.05), and significantly higher EPM open arm entry times and stay time percentage (P<0.05).

  Conclusion The BXD, 5-HT3R specific agonist 1-phenylbiguanide and CB1R specific antagonist Rimonabant can effectively correct the anxiety like behavior of rats with PMDD anxiety disorder.
