[Animal modeling] - Effect of Sishen Pill on the expression of TLR4 and NOD2 in colon tissue of rats with ulcerative colitis

  Objective To compare the changes of Toll-like receptor 4 (TLR4) and nucleotide-binding oligomeric domain protein 2 (NOD2) in the colon tissue of rats before and after treatment, and to explore the mechanism of Sishen pill in treating ulcerative colitis (UC) from the perspective of repairing the intestinal mucosal barrier.

  Methods The rat model of UC was prepared by enema with trinitrobenzene sulfonic acid/ethanol solution. Forty rats were randomly divided into blank group, model group and sulfasalazine group (0 36 g/(kg · d) and traditional Chinese medicine group (5 g/(kg · d)), the gene and protein expression of TLR4 and NOD2 in colon tissue were observed by HE staining, RT-qPCR, SP method and Western Blot method.

  Results Compared with the blank group, the colon injury in the model group was serious and the injury score was significantly higher (P<0. 05) 01), TLR4, NOD2 mRNA and protein expression were significantly increased (P<0 01,P < 0. 05) ; Compared with the model group, the colon injury of rats in each treatment group recovered to a certain extent, and the injury score was significantly reduced (P<0 01,P < 0. 05), the expression of TLR4 and NOD2 mRNA and protein were significantly decreased (P<0 01,P < 0. 05) 。

  Conclusion Sishen Pill can improve the immune function of rats, inhibit intestinal inflammatory reaction, and achieve the goal of treating UC.
