[Animal modeling] - Cannabinol reduces the changes of monoamine neurotransmitter induced by methamphetamine in rats

  Objective To explore the effect of cannabinol on monoamine neurotransmitters in the central nervous system in methamphetamine dependent rats, explain the therapeutic mechanism of CBD on METH dependence, and provide reference for the development of drug dependence abstinence drugs.

  Methods METH-dependent rat model was established by repeated intraperitoneal injection of METH (2 mg/kg) and conditioned location preference; Five brain regions related to the reward system, including the nucleus accumbens, the frontal cortex, the ventral tegmentum of the midbrain, the striatum and the hippocampus, were extracted. Three monoamine neurotransmitters, dopamine, serotonin and norepinephrine, were separated by HPLC; Quantitative analysis of three monoamine neurotransmitter levels by mass spectrometry.

  Results Repeated administration of METH (2 mg/kg) significantly up-regulated the level of DA and down-regulated the level of 5-HT in NAc, FC, VTA, CPU and Hip of rats, accompanied by the up-regulation of NE in NAc and VTA; The intervention before CBD (10, 20, 40, 80 mg/kg) can weaken the effect of METH on the monoamine neurotransmitters in a dose-dependent manner.

  Conclusion CBD may weaken the reward effect caused by METH dependence by maintaining the steady level of monoamine neurotransmitters in the reward brain area of rats.
