[Animal experiment]-Protective effect of Apelin-13 on focal cerebral ischemia-reperfusion injury in rats

Objective: To study the protective effect of apelin-13 on focal cerebral ischemia-reperfusion injury in rats and its possible mechanism.

Method: An improved thread bolus method to establish a local CIRI model of SD rats. Sham operation group, model group, low-dose Apelin-13 intervention group A, medium-dose group B, high-dose group C, 5 groups, were injected into the lateral ventricle of Apelin-13, and scored neurological function, cerebral edema, brain kinase 1/2 (ERK1/2) can measure infarct volume, observe cell apoptosis and detect brain tissue malondialdehyde (MDA), superoxide dismutase (SOD) activity and extracellular regulatory proteins.

Results: (1) The neurological scores of groups B and C were lower than those of the model group (P0.05). The expression of p-ERK1/2 protein in the model group and intervention group was higher in the sham operation group (P\u003c0 .05); The intervention group was higher than the model group (P\u003c0.05).

Conclusion: Apelin 13 may have a protective effect on focal CIRI in rats by inhibiting oxidative stress; ERK1/2 signaling pathway may be involved in the protective mechanism of Apelin 13.
