OBJECTIVE: To study the inhibitory effect of Biejiajianwan on the growth of human hepatocellular carcinoma HepG2 transplanted tumor in nude mice and the effect of Wnt/β-catenin signaling pathway related protein expression, and to explore the molecular mechanism of Bjiajianwan on hepatocellular carcinoma.
Methods: Nude mouse tumor formation experiment, to observe the inhibitory effect of Biejia Jianwan on the growth of human liver cancer HepG2 transplanted tumor in nude mice, immunohistochemical method to detect the expression level of PCNA in human liver cancer HepG2 tumor tissue transplanted in nude mice, TUNEL method to detect human The tumor cells of hepatocellular carcinoma HepG2 transplanted nude mice were apoptosis, and the expression levels of β-catenin and Tbx3 were detected in the tumor tissues of human hepatocellular carcinoma HepG2 transplanted by Western blotting.
Results: B Jia Jianwan can significantly inhibit the growth of human liver cancer HepG2 transplanted nude mice (P\u003c0.05). Negative control (NC) group (5.5±0.90), high-dose (H) group (22.9±1.22), medium-dose (M) group (14.7±0.50) significantly increased the apoptosis rate of transplanted tumor cells (P\u003c0 .05); H (40.9±2.31), M (53.5±2.41), low-dose (L) group (62.3±1.80) compared with NC group (93.5±1.70), the proportion of PCNA positive cells was significantly higher. Has been reduced (P\u003c0.05). At the same time, Biejiajian Pills can inhibit the signaling molecules β-catenin and Tbx3 in the Wnt /β-catenin signaling pathway.
Conclusion: The anti-liver cancer mechanism of Biejiajianwan may be related to the inhibition of PCNA expression and the regulation of the expression of related proteins in the Wnt /β-catenin signaling pathway.
