OBJECTIVE: To study the expression of cannabinoid CB1 receptor in the striatum of rats with Parkinson's disease (PD) dyskinesia (LID) and the intervention effect of Compound Dihuang Decoction.
Method: 6-hydroxydopamine (6)-OHDA) damages the substantia nigra laterally to prepare a rat model of Parkinson's disease. L-dopa + benzozirasid (50 mg/50 mg/50) is used to prepare LID Rat model. They were intraperitoneally injected with kilograms of levodopa and 12.5 mg/kg benserazide, and they were divided into a random LID group and a combined dihuang formula group. In addition, 6 rats were prepared for each group, and rats in the normal control group and the sham operation group were used as the control group. After 4 and 6 weeks of neurobehavioral testing, the rats were sacrificed and the striatum was collected. The expression of the cannabinoid CB1 receptor in the striatum of each group of rats was determined by Western blotting.
Results: Increasing the modeling time can increase the AIM score of LID rats (P0.05), increase the turnaround time (P0.05), and combined with Dihuang Decoction can improve the above changes. The expression of receptors increased with the increase of modeling time, and showed a downward trend (P\u003c0.01).
Conclusion: The content of cannabinoid CB1 receptors in LID model rats is significantly increased, and there are changes such as yin deficiency and wind. The complex intervention of Dihuang Decoction in LID model rats can better reflect the severity of the syndrome, activate the stria cannabinoid CB1 receptor, inhibit the release of excitatory amino acids (mainly glutamate), Intracellular cascades may trigger a response to weaken the excitability of neurons. Reduce the excitotoxicity of levodopa.
