OBJECTIVE: To investigate the effect of Jiuxu Xiening Granules on the gene expression of TLR4 and NF-κBp65 protein in the colon tissue of rats with ulcerative colitis with spleen and kidney yang deficiency.
Method: Use cortisone and TNBS (2,4,6-trinitrobenzene sulfonic acid) mixed with ethanol enema to establish an animal model. Using immunohistochemistry SP method and RT-qPCR method, 90 rats were randomly divided into blank group, model group and Jiux Ling granule treatment 7, 14, 21 d group and positive control (SASP) group. We detected TLR4, NF-κBp65 protein and gene expression in the rat colon tissue after treatment.
Results: The colonic mucosal morphology score results showed that the scores of each treatment group were reduced, and the difference was statistically significant (P\u003c0.01). The results of immunohistochemical SP method showed that the expression of TLR4 and NF-κBp65 in the model group increased significantly (P\u003c0.01); after drug intervention, the expression of TLR4 and NF increased in each treatment group-the expression of κBp65 decreased (P \ u003cu\→ u003c0.05); T-qPCR results showed that, compared with the blank group, the expression of TLR4 and NF-κBp65 in the model group increased (P \u003cu\→ u0.01c0.01). Compared with the model group, the expressions of TLR4 and NF-κBp65 in each group were reduced after treatment (P\u003c0.05).
Conclusion: The flowing particles can reduce the inflammation of the colonic mucosa and play a role in mucosal repair. The reason is related to the decreased transcription and protein expression of TLR4 and NF-κBp65 genes in the colon tissue of the rat model of UC with insufficient spleen and kidney.
